Introduction and Application
Tyrosinase plays a key role in melanin production because it controls the rate of melanin deposition. 4-Butylresorcinol has an inhibitory effect on melanin production by directly inhibiting tyrosinase activity, as well as inhibiting the synthesis of tyrosinase in B16 melanoma cells without causing any cytotoxicity. In some in vitro studies, 4-butylresorcinol has been shown to inhibit melanin production, as well as tyrosinase activity and TRP-1.
Mechanism of action:
1 Before melanin synthesis, it interferes with the synthesis and glycosylation of tyrosinase, preventing the enzyme from being absorbed by melanosomes
2 During melanin synthesis, it inhibits the activity of the enzyme, acting as a competitive inhibitor of tyrosinase and TRP1 enzymes, reducing the formation of byproducts that promote melanin production.
3 After melanin synthesis, it increases tyrosinase degradation, inhibits the transfer of melanosomes to keratinocytes, and has a light-stripping effect due to the action of fatty acids. This function is enhanced by the presence of fatty acids and regulates the degradation of tyrosinase.
