Introduction
Bis(2-mercaptoethyl) sulfone(BMS) is a water-soluble reagent that can be used to reduce native disulfide bonds in proteins. It is considered a superior reducing agent to DTT. It has been shown that under non-denaturing conditions, the reduction rate of accessible disulfide bonds in immunoglobulins and trypsinogen is 5 to 7 times that of DTT at pH 7. At pH 7, BMS reduces the relatively inaccessible disulfide bonds in α-chymotrypsinogen A 2.3 times faster than DTT. The pKas are 7.9 and 9.0, respectively.
