Introduction
AZD6738 is a potent ATR kinase inhibitor with an IC50 of 1 nM for the isolated enzyme and 74 nM for ATR kinase-dependent CHK1 phosphorylation in cells.
It has potential antitumor activity.
Application
ATR kinase inhibitors selectively inhibit ATR activity by blocking downstream phosphorylation of the serine/threonine protein kinase CHK1.
This blocks ATR-mediated signaling and results in inhibition of DNA damage checkpoint activation, disruption of DNA damage repair, and induction of apoptosis in tumor cells.
