Introduction
X-GAL, chemically known as 5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside, is a substrate for β-glucuronidase, which converts it into an intense blue tryptophan. It is used to detect Gus genes in bacterial colonies, such as Escherichia coli, or for histochemical applications.
X-GAL (CAS 7240-90-6) is a white to off-white crystalline powder with a melting point of 225–230°C, density 1.9 g/cm³, and is soluble in organic solvents such as dimethylformamide and methanol. As a chromogenic substrate for β-galactosidase, it produces a blue product under enzymatic catalysis. It is primarily used in blue-white screening assays and for detecting β-galactosidase activity.
