Introduction and Application
1-Adamantanol, chemical formula C₁₀ H₁₆ O, CAS number 768-95-6. This substance is a white crystalline powder with a melting point of 247℃ (sublimation) and a density of 1.2 g/cm³. It is soluble in organic solvents such as methanol and ethanol, insoluble in water, and has sublimation properties.
1-Adamantanol is used to make synthetic adamantane derivatives and adabalone.
Application
1-Adamantanol is used for the synthesis of adamantane derivatives and pregabalin. Reacting with vinylidene chloride yields 1-adamantaneacetic acid. 1-adamantaneacetic acid is further brominated and hydrolyzed to 3-hydroxy-1-adamantaneacetic acid and then reacted with vinylidene chloride to obtain 1,3-adamantaneacetic acid.
The Friedel-Crafts alkylation reaction between 1-adamantanol and acetanilide occurs at room temperature in a 98% sulfuric acid medium, and 1,3-di(4-acetaminophenyl)adamantane can be directly synthesized. 1,3-di(4-aniline)adamantane can be synthesized by alkaline hydrolysis, which has the advantages of high reaction yield and easy purification, greatly facilitating the synthesis of 1,3-adamantane diaryl derivatives
